Simultaneous determination of three traditional and two novel Antiepileptic Drugs using Micellar Liquid Chromatography

Abuse of antiepileptic drugs (AED) in young generation is a rising problem in India. The problem is grave when these AEDs are taken in combination with other drugs or second or third generation AED. AEDs are now also been related with increasing suicidal tendency, thus to detect these drugs in biological fluid is an important task. By mean of this research work, efforts have been made to detect combination of old and new AEDs in biological fluid using simple micellar liquid chromatographic method with photodiode array detection. This paper discuss the determination of five antiepileptic drugs (carbamazipine, clobazam, lamotigine, phenytoin and topiramate) using C18 column (5μm, 250×4.6mm) hybrid mobile phases containing sodium dodecyl sulfate (SDS) as surfactant and pentanol as modifier. Detection was performed with a diode array detector at 230nm. After the application of an interpretative strategy for the selection of the optimum mobile phase, all novel and conventional AEDs were resolved and determined in serum samples by direct injection, using a mobile phase made up of 0.06M SDS 6%(v/v) pentanol, pH 5 and an increased flow (1ml/min flow rate was carried out for the analysis up to 12 minutes and after it flow-rate was increased in steps to 0.25 ml/min. till the flow reached 1.5 ml/min.) with an analysis of < 17 min. Calibration was linear in the range 0.005μg/ml to 10 μg/ml with r>0.9997 for all AEDs. The statistical evaluation of the method was examined by performing intra-day (n=5) and inter-day (n=5) study and the result was found to be satisfactory, with highly accurate and precise results. © 2013 Universal Research Publications.
